Examples of Poor Arrays 1) This array is generally acceptable, but it has areas of non-specific hybridization where it has been scraped in the upper-right corner and across all of the lower 10 rows. These areas should be flagged before analysis. 2) This array dried out before being washed thoroughly, which caused high overall background,and characteristic cloudy regions where the radioactive target solution has precipitated on the array. Also note that the array was moved during the exposure on the phosphorimager screen, which gives it a blurry appearance. 3) This array is probably too light to analyze. This could be caused by low specific activity probe or wash conditions that are too stringent. The array should be exposed for a longer period or a new target should be prepared with higher specific activity. 4) This last array is an example of an array that has high background evenly across the entire array and should be washed again in more stringent conditions before analysis. |
