Novel Vaccine Against Colorectal
Carcinoma (CRC)

researchers are investigating novel approaches for immunotherapy
of cancer. For one such approach, a replication deficient adenovirus
expressing the CRC associated antigen GA733 2 was constructed for
active immunotherapy of CRC patients. In Phase II trials, an immune
response to this antigen (induced by administration of a monoclonal
antibody to GA733 2 antigen) was effective in preventing recurrence
of CRC. Administration of GA733 2 inhibits growth of established
CRC cells in mice. Since this effect may be mediated by cytotoxic
antibodies, and/or cytolytic and proliferative T lymphocytes, active
immunotherapy using the GA733-2 antigen may be more effective than
passive immunotherapy.

Applications and Advantages:
The adenovirus expressing the GA733-2 antigen has potential
as a vaccine for CRC patients. A Phase I trial using this technology
is planned.
