How to Obtain Services

When beginning a project, it is important to first consider the immonogenicity of the protein.In general, the facility suggests adding a GST tag to the protein to increase its immunogenicity. The facility will perform fusions with proteins that are not tagged, however, proteins with GST tags have been much more successful compared to non-tagged or 6-his-tagged proteins. Any specific questions about this can be directed to either Andrew Caton or Julia Conicello.

Instructions for beginning a project:

1) 500 ug of antigen in PBS at a concentration between 0.5 and 2 mg/ml is required for injections and screening.

2) If the protein is a fusion protein (e.g. GST), ~100 ug of the purified fusion protein backbone (e.g. GST alone) is required to use as a negative control during screening.

3) A request form must be completed. (The request form provides a space for an NCI grant number.  If the project is supported by an NCI grant, the grant number is required.  If the project is supported by another source, no number is required.)

4) Indicate whether the project is to be done with mice or rats.  Mice are used most often.  Since the facility uses a mouse myeloma line as a fusion partner, the resultant hybridomas from a rat fusion are unstable due to the cross-species pairing of cells. These hybridomas are much harder to maintain than those from a mouse fusion. Therefore, mice are generally recommended except in special cases. Related questions should be directed to Andrew Caton.

5) Anyone who does not work directly for the Wistar and cannot place orders through the Wistar Accounting system (including Affiliate Cancer Center Members) must provide a PO for the cost of services before any work can be initiated.
